Instructions1. Prepare working Iodide Reagent 2: (a) ﬁll empty plastic vial to thread level with deionized (DI) or distilled water (not supplied); reverse osmosis (RO) water may not be sufﬁciently pure; add 1 drop Iodide Reagent 2a (using dispensing pipette) and 1 drop Iodide Reagent 2b; (b) cap vial and mix; (c) wait 5 minutes before using. The working Iodide Reagent 2 solution is usable for 3–4 hours.2. Using supplied spoon, dispense one level measure of Iodide Reagent 1 to a test cavity.3. Add one full stem of sample using sample pipette to the test cavity containing Iodide Reagent 1. [If a blue color develops now, elemental iodine is indicated. This should be viewed as a hazardous status. Use a dechlorinator such as Prime™ to remove elemental iodine.]4. Add 2 drops Iodide Reagent 2 (that you already prepared in Step 1 above) with dispensing pipette to the test cavity and stir brieﬂy with straight end of spoon. It is normal for not all of the powder to dissolve.5. Promptly compare color to color chart as soon as color peaks (20–40 seconds). Peak color will remain stable for about a minute. Rinse out test plate after use and dry.
Reference testThe proper performance of this kit may be validated by running a test in the normal manner except that the reference sample is used in place of a aquarium sample. It is not necessary to run a reference test to use this kit. The only time you might choose to run a reference test is if you have cause to believe the test is giving incorrect results. If you run a test using the reference sample and obtain the correct result (based on the known reference value provided) then you know the test is giving correct results. To run a reference test, use the Iodide Reference as the sample in a iodide test. Reference value is 0.06 mg/L.
Hints: If color is very faint you can increase the sensitivity of the test by increasing the number of drops used in Step 4. For example, you can triple the sensitivity by using 6 drops (e.g. a value of 0.02 mg/L will read as 0.06 mg/L). Likewise, if color is off scale you can decrease sensitivity to get the color back on scale by either (a) reducing the # of drops in Step 4 or (b) by prediluting your sample, or by a combination of both. Predilution can be done in the test well. A full stem of the sample pipette is equal to 14 drops, so adding 7 drops of sample and 7 drops of DI water will yield a 50% dilution (e.g. a 0.1 mg/L sample diluted will read as 0.05 mg/L). Better accuracy will be attained with colors corresponding from low to the mid-range of the color scale. Chlorine and chlorine removing products will interfere. If a water change has been made with water containing chlorine or a chlorine removing product, wait about 8 hours before testing. Neither chloride nor bromide interfere. Ozone will likely interfere. The use of ozone is not recommended in seawater, since it generates toxic free halides (chlorine, iodine, bromine).
Interpretation: This kit is speciﬁc for iodine/iodide and easily detects down to 0.001 mg/L. The color scale peaks at 0.10 mg/L. Natural seawater (NSW) concentration of iodide is 0.06 mg/L. Ideally, it is advisable to maintain iodide close to NSW concentration. Concentrations above NSW may not be harmful, and, within limits, may even be beneﬁcial, but maintenance at elevated concentrations should be considered experimental. If you ﬁnd that your reef tank has low iodide, this may be a chronic status and concentration should be raised gradually, since a sudden increase in iodide may be detrimental.
